Black cumin seed extract

Description

Black cumin seed (Nigella sativa ) is an annual herbaceous plant and a member of the Ranunculaceae (buttercup) family. The fruit of the plant, the black seeds, accounts for its name. Black cumin seed (also called black seed) should not be confused with the herb, cumin (Cumunum cyminum, which is found in many grocery stores.



Considered native to the Mediterranean region, black cumin seed is cultivated in North Africa, Asia, and southeastern Europe. The largest producers of black cumin seed are Egypt, India, Pakistan, Iran, Iraq, and Turkey. Other species, such as Turkish black cumin (Nigella damascena ), are not used medicinally; and one type, Nigella garidella, is even poisonous.



Playfully referred to as "Love in the Mist," the black cumin seed plant has leaves that grow in pairs. The lower leaves are short and supported by slender stems, while the upper leaves generally grow to approximately 4 inches (10 cm) in length. The stalk of the plant, with its bluish white flower petals, can grow up to 18 inches (46 cm) in height while its fruit matures. At first, the seeds (the fruit of the plant) are held in a capsule in the center of the flower. The capsule opens upon maturity, revealing lightly colored seeds. It is only upon their exposure to air that the seeds become black.



Most often, the extract is produced by a process referred to as cold pressing. Temperatures no higher than 140–176°F (60–80°C) are applied to the seeds to help release the oil and preserve its benefits.



Rich with compounds such as nigellone and thymoquinone, black cumin seed is thought to contain over 100 ingredients; many remain unknown. However, experts agree that the most important compounds contained in the extract are the fatty acids and nutrients. Some components of black cumin seed extract are as follows:

General use

Black cumin seed has been used for centuries to treat respiratory and digestive problems, parasites, and inflammation. In ancient times, it was a remedy for a variety of health conditions including, colds, infections , headaches, and toothaches. The pharoahs' personal doctors are reported to have offered black cumin seed as a digestive aid after large meals. In fact, the extract was found in the tomb of King Tutankhamun, presumably to protect him in the afterlife.



Black cumin was also used as a remedy for skin diseases, dry skin, dandruff , and wounds .



At one time, black cumin seed was highly valued in Europe, but by the eighteenth century it had lost popularity, and was primarily used as a garden decoration. However, black cumin seed extract has regained popularity, and is now more widely used as a remedy in Europe and North America.



Many herbalists in current times embrace the healing properties of black cumin seed extract. For example, the extract is sometimes used externally to treat such skin care problems as psoriasis, eczema , and dry skin, and internally to treat stomach problems, respiratory ailments, and allergies , as well as to improve circulation and the immune system. In recent years, the extract has been the subject of immune system research.



One reason that is often given for the medicinal value of black cumin seed extract is its richness in polyunsaturated fatty acids, which help to produce prostaglandin E1. Prostaglandin E1 has many functions in the body, particularly in relation to the immune system, sugar metabolism, skin infections, and blood clots . It is also believed to protect the stomach lining.



Experts point out that the medicinal value may be provided by a unique and mysterious synergy (combined action) between the multitude of compounds present in the seeds. In addition, the extract, which is more concentrated than the seeds alone, is said to have greater healing power. A study at Cairo University in Eqypt showed a boost in antibacterial activity when the extract was used in combination with antibiotics such as streptomycin and gentamicin. In the same study, it showed additional antibacterial function in combination with erythromycin, tobramycin, doxycycline, and ampicillin, to kill E. Coli and the pathogenic yeast, Candida albicans. In addition, the study showed that the extract destroyed non-fatal subcutaneous staphylococcal infection in mice.



In 2003, one study noted the antifungal activity of black cumin seed extract against Candida albicans. In the study, mice were injected with Candida albicans, producing colonies of the organism in their liver, spleen, and kidneys. The researchers found that treatment with black cumin seed extract 24 hours after inoculation inhibited growth of the Candida albicans. With continued treatment, the extract significantly decreased the amount of Candida albicans found in the kidneys, liver, and spleen.



Aside from verifying its antibacterial and antifungal properties, researchers in recent years have tested the anti-inflammatory and analgesic effects of black cumin seed extract. In 1995, a group of scientists from the Department of Pharmacy at King's College in London found that the extract contains these properties, and is an antioxidant as well. They believe the anti-inflammatory and antioxidant abilities may be linked to ingredients such as thymoquinone and unsaturated fatty acids. Ultimately, the researchers concluded that black cumin seed extract is a justified treatment for rheumatism and related inflammatory diseases.



In 2001, a study performed at the Department of Pharmacology at King Faisal University in Saudi Arabia, reported anti-inflammatory and analgesic activity from the use of black cumin seed extract in animals. Paw edema (swelling) was reduced, as was reaction time in response to extreme heat. A 2003 study confirmed the analgesic effects of the extract. Studies in this area are likely to continue well into the future.



Researchers have also investigated and verified the extract's antihistamine activity, focusing on nigellone, an ingredient in black cumin seed extract. One 1993 study found that nigellone acted as an inhibitory agent on histamine (a substance involved in an allergic response, causing widening of blood vessels and tightening of bronchial passages) by inhibiting protein kinase C, known to initiate histamine release. In 2003, another study concluded that black seed oil is an effective treatment for allergies.



Preparations

There are many applications made with black cumin seed extract. It can be found in teas, cough syrups, wound salves, compresses, massage oils, and other products. Black seed honey, soap, shampoo, and creams are all available commercially.



The extract has a strong flavor, which is improved by mixing it with honey. Herbal teas also help dilute its strength. As with any product used for medicinal purposes, it is important to read and follow the label instructions and warnings.



Although black cumin seed extract is not normally associated with severe skin irritation, a skin patch test should be conducted before using it for the first time. A small amount of diluted extract is placed on the inside of one elbow and covered with a bandage. After 24 hours, any redness or irritation is indicative of a negative reaction. This test should be done before a person proceeds with more extensive use.



Black cumin seed extract, in these dosages, is used as a remedy for the following conditions:



Headache. A few drops of the diluted extract are rubbed on the patient's forehead. Some patients may also find it helpful to take 1/2 teaspoon of the extract after breakfast, lunch, and dinner.

Cough. The dose is 1/2 teaspoon of diluted black cumin seed extract in the morning. A dry cough may require one teaspoon of the extract twice a day, mixed with one cup of coffee or hot tea. The extract can be rubbed on the chest and back for additional relief.

Common cold. One teaspoon of the extract is mixed with hot lemon tea and honey two or three times a day.

Diarrhea. One teaspoon of extract is mixed with one cup of yogurt twice a day.

Precautions

Black cumin seed extract is not to be used during pregnancy .



Its safety in young children has not been established. Patients with liver or kidney disease are advised not to use this product unless a physician directs them to do so.



Black cumin seed extract is said to lower blood sugar levels; therefore, a diabetic patient is advised to consult with a physician before using.



Side effects

In general, if used as directed, black cumin seed extract is not associated with serious side effects. However, it has been reported that black cumin seed extract has a very low degree of toxicity, and may cause significant negative effects on liver and kidney function. A recommended daily allowance (RDA) has not been established for the extract, so it is wise to consult with a physician before beginning any internal treatment.



Interactions

There does not appear to be a list of serious interactions associated with the use of black cumin seed extract; however, it is recommended that anyone taking prescription drugs seek the opinion of a physician and/or pharmacist before using black cumin seed extract in combination with the prescribed treatment.

An alternative treatment for Candida infections with Nigella sativa extracts

Abstract

Background Nigella sativa is a herb from the Mediterranean region with antidiabetic, bronchodilator, antioxidant, hepatoprotective, lipid lowering, anti-inflammatory and analgesic properties.



Purpose This study aimed to reveal the antifungal activity of aqueous, methanolic and chloroform extracts obtained from the plant seeds, compared with the effect of traditional antifungals.



Materials and methods Using standard mycological diagnostic methodology The authors isolated and identified 20 strains of Candida albicans from pathological products collected from patients hospitalised in different departments of the Craiova Emergency Hospital. Aqueous, methanolic and chloroform extracts were made from the seeds of Nigella sativa, in decreasing dilutions, in which Wattman filter paper discs were soaked and dried and then used to achieve the antifungal graph by using the Kirby-Bauer diffusion technique. Simultaneously, the testing was repeated using standard antifungal disks (Becton Dickinson) and the two sets of results compared. The antifungal effect was assessed by measuring the diameter of the inhibition zone, noting the concentration per disk.



Results The results show that methanolic extracts of Nigella sativa have the strongest antifungal effect followed by the chloroform extracts. Aqueous extracts showed no antifungal activity.



Conclusions The research shows treatment with natural products in a good light as an alternative for treating fungal infections. The authors envisage Nigella sativa extract enhancing the effect of conventional therapy.

Black seed is an effective antiinflammatory and analgesic substance

Animal studies have confirmed the popular claim that black seed is an effective antiinflammatory and analgesic substance. The mechanism by which black seed exerts its antiinflammatory action appears to be as a potent inhibitor of eicosanoid generation, namely thromboxane B 2 and leucotrienes B 4, by inhibiting both cyclooxygenase and lipooxygenase, respectively. In other studies, thymoquinone has been reported to have potent superoxide anion scavenging abilities and to inhibit iron-dependent microsomal lipid peroxidation. This is promising considering the fact that superoxide reacts with protein and non-protein sulfhydryls and polyunsaturated fats and initiates sprecific reactions, thus damaging cells and causing inflammation. Meanwhile free radical oxidative stress is implicated in many inflammatory diseases. Therefore, it is reasonable that the anti-inflammatory activities of thymoquinone are attributed to its antioxidant effects. Interestingly, it was found that the whole oil had both antioxidant and anti-eicosanoid effects greater than thymoquinone, the oil’s active constituent.



A recent study found black seed and thymoquinone may be an effective treatment for rheumatoid arthritis. This comes as no surprise, since it has been shown that thymoquinone is an inhibitor that is more potent than indomethacin of COX-2-catalyzed PGE2 production.



The immuno-modulatory properties of black seed and thymoquinone support its traditional use as a treatment for rheumatism and related inflammatory disorders.

Chronic diseases are the leading causes of death and disability in the United States, advises the Centers for Disease Control and Prevention (CDC).


Chronic diseases are heart disease, stroke, cancer, diabetes, and arthritis and are the most common and costly of all health problems for Americans.

These chronic diseases are also the most preventable, stressing the importance for early detection.

Some believe early detection, like an ounce of prevention, is worth a pound of cure.

One Hernando county physician has implemented a practical approach for local patients to spot small problems before they become big ones.

Dr. Adel M. Eldin, M.D., specializing in internal medicine and cardiology, his practice is Brooksville Cardiology, explained his health and wellness program, which focuses on preventative care. He presented his program to more than 50 participants gathered at Palace Grand in Spring Hill, listening to testimonials from patients who have experienced success with the cardiology center.

"It's possible to detect problems before there's an emergency and 95 percent preventable, given our knowledge, medications and lifestyle modifications," Eldin said.

But there's one problem, Medicare and other insurance companies don't cover preventive screenings without the presence of symptoms, he advised.

"Insurance focuses on illness, rather than wellness," he said. "Wellness is there for you."

"When you invest in your health, you won't have to be part of the insurance jungle out there." Eldin said. "I've spent five years introducing a new program that identifies risks, teaches preventive care and provides an on going partnership in healthy living."

Black seed oil proved to be an effective adjuvant for the treatment of allergic diseases, based on the research.

The seeds/oil has anti-inflammatory, analgesic, antipyretic, antimicrobial and antineoplastic activity. The oil decreases blood pressure and increases respiration, according to other research regarding its pharmacological and toxicological properties.

The seeds have been used in the Middle East to treat asthma, bronchitis, rheumatism and related inflammatory diseases.

Researchers at Kimmel Cancer Center, Thomas Jefferson University in Philadelphia have studied that thymoquinone, an extract of N. sativa seed oil, blocked pancreatic cancer cell growth and killed the cells, acting as a Histone deacetylase inhibitors (HDAC) for use in psychiatry and neurology as mood stabilizers and anti-epileptics

Black Seed Oil and Asthma

Asthma is thought to be caused by a number of environmental and genetic factors and is heavily influenced by the immune system. New research has shown that the oil of a tiny black seed from Western Asia may be the key to both the prevention and treatment of immune-related Asthma.

The Nigella sativa (black seed) oil contains over 100 known health-promoting properties including a wide range of the most potent antioxidants, B vitamins, 15 amino-acids, essential fatty acids, and an array of vital organic minerals. However, researchers believe that it is a substance called ‘nigellone’ that may be the real hero as it protects sufferers from histamine-induced bronchial spasms and significantly relieves the symptoms of asthma, bronchitis, and general coughing.

Twenty-nine asthmatic adults were randomly divided into a control group (14 patients) and treatment group (15 patients), and monitored for 3 months.Asthma symptom score, asthma severity, frequency of symptoms/week and wheezing were recorded on the first visit, then again after 45 days and finally, at the end of the study. Pulmonary function tests (PFTs) were also measured, and the drug regimen of the patients was evaluated at three different visits.
The researchers found that all asthma symptoms, frequency of asthma symptoms/week, chest wheezing, and PFT values in the treatment group significantly improved in the second and third visits compared with the first visit. In addition, further improvement of chest wheezing and severity of disease on the third visit were observed compared with the second visit in this group. Most significantly, by the the third visit (90 days) all symptoms in the treatment group were significantly different from those of the control group.

In addition, the researchers observed that the usage of inhaler and oral ß-agonists, oral corticosteroid, oral theophylline and even inhaler corticosteroid by those patients in the treatment group decreased by the end of the study while there were no obvious changes in usage of the drugs in control patients.

Black Seed Oil For Hemorrhoids?

Black seed oil for hemorrhoids sufferers? There are those who claim that black seed oil will cure any ailment from cancer to flat feet. A miracle cure? I decided to investigate and see what use black seed oil would be for hemorrhoids sufferers in particular. In my researches I started with Wikipedia and ended up at the British Library for nearly three weeks. This is what I found about the healing properties (or otherwise) of black seed oil for hemorrhoids and other ailments…

Black seed oil comes from an annual flowering plant called nigella sativa, which is native to south and south-west Asia. Its fruit is a large inflated capsule, full of seeds. These black seeds are used as a spice and are often called kalon ji – or black onion seeds. There are many names and it is sometimes confused with cumin, sesame, fenugreek and other aromatic seeds. The oil that is obtained from these seeds is said to have miraculous healing powers, especially in Islamic cultures. The sayings of Prophet Mohammed underline the significance of black onion seeds. Effects of Nigella Sativa and Albendazole alone and in combination in Toxocara Canis infected mice

Abstract

Objective: Nigella sativa extract has been used in the Middle East as a traditional medicine for several complaints. We aimed to evaluate the biochemical, histopathological and hematologic changes in Toxocara canis-infected mice after treatment with Nigella sativa extract, albendazole or a combination of both.

Methods: This comparative study was conducted between June and July 2008 at the Itarran University Saniturla Turkey. Sixty healthy adult BALB/c male mice were randomly divided into six groups (D0-D5). Mice in groups D1-D5 received 500 embryonated T. canis eggs via esophageal tube. Groups D0 and D1 served as a non-infected sham group and an infected control group, respectively. Groups D2 and D3 received 100 and 200 mg/kg N. sativa extract (NSE), respectively. Group D4 received 100 mg/kg albendazole. Group D5 received the combination dose (100+100 mg/kg NSE+albendazole).

Results: Treatment with N. sativa of both doses or the combination dose of N. sativa and albendazole reduced the degree of inflammation and necrosis, lead to a reduction in the percentage of eosinophils and decreased the elevated liver enzyme levels.

Conclusion: These results indicate that N. sativa has a potent effect in protection against organ damage induced by T. canis infection.

Introduction
Herbal medicines are used for treatment of many clinical diseases, but little is known about their mode of action. In recent years, Nigella sativa L (Family Ranunculaceae) has been used in the Middle East as a traditional medicine for many complaints.1 The majority of research has focused on its antifungal, antibacterial, antihelminthic, antioxidant and hepatoprotective effects.2-6 Toxocariasis is a zoonotic disease caused by ingestion of embryonated eggs of the roundworm Toxocara canis of which the adult form inhabits the small intestine of infected dogs. About 100% of puppies have ascarid infections during the first 3 weeks of life and 50% at 4 months. During this period, puppies excrete billions of ascarid eggs.7 Mammals, including rabbits, rats, mice and monkeys, serve as paratenic hosts. Humans are infected accidentally by ingestion of the embryonated eggs, resulting in visceral larva migrans (VLM) or ocular larva migrans (OLM).8 Experimentally, mice infection results from ingestion of the second stage of the larvae.9 The second stage larvae hatch in the stomach of the mouse and invade whole small intestine within 2 hours.10 The larvae reach the liver and lungs within the first 2-3 days, then the muscular system and brain 7 days after infection. The larvae produce antigen, enzymes and cuticle components which eventually cause tissue damage, necrosis and inflammatory reactions that lead to an increase in eosinophil count.11

No specific drug for the treatment of VLM and OLM has been investigated in well-controlled studies.3 However, a number of other studies concerning the effects of several antihelminthic drugs on human or experimental animal models have been conducted, using different therapeutic procedures. The drugs available for the treatment of toxocariasis presenting as VLM is limited to albendazole, tiabendazole, tinidazole and diethylcarbamazine.12,13

Considering the side effects of these drugs we focused on traditional medicine Nigella sativa with the aim to evaluate the biochemical, histopathological and haematologic changes in T. canis-infected mice after treatment with N. sativa, albendazole or a combination of both.


Methods

Animals Design:

A total of 60 healthy adult male BALB/c mice weighing 32 ± 2 g were maintained in accordance with the National Institutes of Health Guide for the Care and Use of Laboratory Animals. Animals were obtained from Istanbul University, Faculty of Science, Department of Biology. Animals were housed individually in polypropylene cages under environmentally controlled conditions (room temperature 22 ± 2°C, humidity 60% and 12/12 light/dark cycle) with free access to standard rodent food (Ankara pellet feed factory, Ankara, Turkey) and water for duration of the study. The Institutional Animal Ethics Committee of Harran University approved animal protocols in advance before the experiment begun. The mice were acclimatized to the laboratory conditions 2 weeks prior to the experiment.

Egg Collection and Preparation:

T. canis worms were collected from naturally infected puppies (taken from a dog shelter in Sanliurfa). The embryonated eggs were removed by dissection of the uteri of the worms according to a method described by Bowman et al.14 The eggs were diluted with 10% gum arabic suspension to achieve 500 embryonated eggs in 1 ml of suspension.

Plant Material and Drug Preparation:

N. sativa seeds were purchased from a local herbal shop in Sanliurfa Turkey. The seeds were cleaned, washed, dried and grounded into fine powder using an electrical mill. The powder was macerated in methanol for 24 hours and then evaporated in a rotary evaporator and dried by lyophilization. Two different doses of extract were prepared (100 and 200 mg/kg bodyweight) and suspended in 10% gum arabic (Merck, Chemical Company, Darmstadt, Germany). Albendazole was purchased from Sigma (St. Louis, MO, USA) and dissolved in 1% dimethyl sulfoxide (DMSO), prepared as 100 mg/kg bodyweight and suspended in 10% gum arabic.

Experimental Conditions:

A total of 60 healthy adult BALB/c male mice were randomly divided into six groups (n=10). Groups D0 and D1 received orally 1 ml of vehicle (10% gum arabic suspension) and served as a non-infected sham group and an infected control group, respectively. Mice in groups D1-D5 received 500 embryonated T. canis eggs via esophageal tube. The treatment was given at 24 hours post-infection in groups D2-D5. Groups D2 and D3 received 100 and 200 mg/kg bodyweight N. sativa extract, respectively. Group D4 received 100 mg/kg bodyweight albendazole. Group D5 received the combination dose (100 mg/kg bodyweight N. sativa extract+100 mg/kg bodyweight albendazole). The treatment was given for 7 consecutive days to all groups.

At the end of the experiment, all animals were killed by cervical dislocation, and their liver, lungs, brain and muscles were removed. Each liver was weighed on an analytical balance and divided into two portions; one portion was used for histopathological examination.

Worm Recovery and Larvae Count:

The left lung, one portion of liver and muscles were finely minced. The larvae were recovered according to the method described by Parsons and Grieve15 with little modification. The tissues were weighed and then digested individually in pepsin-HCl solution (pH 1-2, 500 IU pepsin/g tissue) (Sigma-Aldrich, Hyderabad, India) and incubated for 3 hours at 37°C with periodic agitation. Cellular debris was removed after centrifugation. Subsequently, the sedimentation of particles in liquid were poured into a centrifuge tube and stored in a refrigerator at 4°C for 24 hours. The sediment liquid was centrifuged for 2 minutes at 1500 rpm and 2 ml of the sediment was collected and vortexed. Larvae in the sediment were counted under a light stereomicroscope. The brain of each mouse was removed, minced into small pieces, macerated carefully between two slides to ensure a thin layer, and the larvae were counted under a light microscope.

Histological Examination:

Organs, including the liver and lungs, were removed, washed with physiologic solution and fixed separately in neutral formaldehyde overnight, dehydrated in graded ethanol, cleared in xylene, and processed in ParaplastR (Fluka Chemika AG Buchs, Switzerland) at 58°C. 4-5 µm-thick sections were prepared and stained with haematoxylin and eosin (H&E). Three slides were prepared for each tissue and randomly selected for examination by a blinded pathologist. The gross necrotic lesions were histologically identified as large, subcapsular eosinophilic areas of necrotic hepatocytes. Inflammatory foci were defined as discrete clusters of 10 to 80 individual, small, nucleated cells visible throughout the liver. The site of inflammation was taken into consideration only if well-defined lesions and inflammatory foci were present. The degree of inflammation or damage was scored and categorized into four levels: normal (0), no inflammation; mild (1+), <3 foci/area; moderate (2+), 3-6 foci/area and severe (3+), >6 foci/area. Three slides were evaluated for each animal. The mean of all the individual scores was calculated and used for analysis.

Serum Enzyme Analysis:

Blood was collected from all animals under ether anaesthesia, and by heart puncturing according to the procedure approved by the Institutional Animal Care and Use Committee (IACUC). The blood samples were allowed to clot at room temperature. Serum was separated by centrifugation at 3000 rpm for 10-15 minutes and utilized for the measurement of biochemical parameters. Assay kits for the measurement of serum enzymes aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) were purchased from Randox Laboratories Ltd (Ardmore, Crumlin, UK). Measurements were performed with a UV spectrophotometer in accordance with the manufacturer\'s instructions.

Statistical Analysis:

Inter-group comparisons were conducted using the Kruskal-Wallis test followed by Dunn\'s multiple comparison of means test. All results are presented as mean±SD. P < 0.05 was considered statistically significant.


Results

The total larval burden in the infected control group D1 was 61. Burden was 28 in the examined lungs (45.9%), 18 in liver (29.5%), 12 in the carcass (19.7%) and 3 in brain (4.9%). Treatment with N. sativa alone reduced the total larval burden to 42 and 37 in groups D2 and D3, which indicated a percentage reduction of 31.1% and 39.3%, respectively, compared with the control group D1 (P<0.05). On the other hand, in group D4 treatment with albendazole alone reduced larval burden to 18, which indicated a percentage reduction of 70.5%. Treatment with the combination of albendazole and N. sativa in group D5 reduced the larval burden to 8, which indicated a percentage reduction of 87%. Significant difference was observed between the treated groups (D4-D5) and the control infected group (D1) (p<0.01).

Percentage of eosinophils in control group was six-fold higher than in sham group. Treatment with N. sativa at doses of 100 and 200 mg/kg reduced the eosinophil percentage from 18% to 7.1% and 7.0%, respectively, while treatment with albendazole alone decreased the percentage of eosinophils to 7.8%. Treatment with the combination of albendazole and N. sativa reduced the percentage of eosinophils to 6.6%.

The histopathological changes observed in the lung and liver sections of the control group compared with the sham group are shown in Figures 1A and B and Figures 2A and B. Histopathological examination revealed inflammatory cell infiltration in the liver (Figure 1A). Severe inflammation and bleeding associated with massive infiltration of inflammatory cells into the interstitial and alveolar tissues was observed in lung parenchyma (Figure 2A). Determination of pathogenicity assessed by inflammatory as well as histopathological changes revealed that T. canis larvae were able to induce varying degrees of histopathological changes in the host organs. Mild (1+) and, moderate (2+) to severe (3+) histopathological changes were observed in the brain, lung and liver of infected mice. Significant difference between the infected control (D1) group and non-infected sham (D0) group were observed with respect to histological changes in brain, lung and liver (p<0.05). Treatment with N. sativa alone decreased the degree of damage and the degree of inflammation in the brain, liver and lung, in a dose-dependent manner.There was significant difference between the N. sativa treated groups (D2-D3) and the control infected group (D1) regarding the histological changes in brain, lung and liver (p<0.05).

Treatment with Albendazole alone also reduced the degree of damage and necrosis compared with T. canis-infected mice tissues.Significant difference between the Albendazole treated group (D4) and infected control (D1) group were observed with respect to histological changes in brain, lung and liver (p<0.05). Moreover, treatment with the combination dose of N. sativa and albendazole reduced the degree of necrosis and inflammation and resulted in a reduction in the percentage of eosinophils (Figures 1 and 2).





There was significant difference between the combination dose of N. sativa and albendazole treated group (D5) and the control infected group (D1) regarding the histological changes in brain, lung and liver (p<0.05).

The results with regard to the effects on serum levels of liver enzymes including ALT, AST and ALP are presented in Table. It was found that infection with T. canis in D1-D5 groups elevated the liver enzymes of ALT, AST and ALP when compared with the non-infected group (D0). A significant increase in serum levels of ALT and AST was found in control infected group (D1) compared with the non-infected group (p <0.01). No significant difference was found between the infected control and non-infected groups regarding serum level of ALP (p >0.05).

Treatment with N. sativa with 100mg/Kg and 200mg/Kg were observed to reduce serum liver enzyme levels as compared to infected control group and albendazole alone treated group Treatment with combination dose markedly prevented the elevation of liver enzymes. Liver enzyme levels of ALT and AST in the treated groups (D2-D5) were significantly lower compared with the infected control group (p <0.05, p<0.05, p<0.05, p<0.05, p<0.01, respectively) (Table).



However, no significant difference was found between the treated groups and infected control group regarding serum level of ALP (p >0.05). Also, there was no significant difference between the treated groups and the non-infected sham group regarding serum levels of ALT, AST and ALP (p >0.05).

Discussion

T. canis is considered to be primarily responsible for VLM and OLM. The route of larval migration in the experimental mouse can be divided into an early hepatopulmonary or visceral phase during the first week after infection, followed by a brain, heart and kidney phase.10 Hepatic damage resulting from T. canis larvae infection is of major importance among liver diseases worldwide.15 Previous work by Lai et al.16 demonstrated that larval antigens are primarily responsible for host inflammatory reactions associated with the infection. The larvae in the liver, elicit T-lymphocyte responses leading to macrophage activation, which seem to play a role in phagocytic removal of unwanted debris at the site of inflammation.16,17 The severity of the disease is determined by the extent of inflammation and amount of larval debris deposited in the tissues.17 In our study, elevated serum levels of AST, ALT and ALP were observed in all infected groups (D1-D5). This seems to be associated with damage of hepatic tissue and impaired cell membrane permeability or is due to deposition of T. canis larvae in tissues. The present study showed reduction of larval burden in treatment groups by both doses of N. sativa (D2, D3) was 31.1% and 39.3%, respectively compared with infected control group (D1). Also, the treatment reduced the degree of damage and inflammation in the liver and the percentage of eosinophils, in comparison to the T. canis-infected control group. The changes observed in the percentage of circulating eosinophils indicated that eosinophil count is an important index of parasite infection and parasitic incidence. Albendazole has been shown to be effective in ocular and visceral larval infections.18,19 The proven efficacy of albendazole in such infections is associated with its nematicidal activity, though the individual immunity has an important role in the response to T. canis infection. Moreover, the hexane extract of C. ambrosioides which is a herbal medicine has been reported to have an anthelmintic activity in vitro by reducing the inflammatory reaction produced by the infection of T. canis larvae in vivo.20

The enhanced liver functions as reflected by a significant decrease in elevated serum levels of the enzymes ALT and AST after treatment with albendazole alone or combination treatment of albendazole and N. Sativa in the present study supports previous studies reporting the efficacy of albendazole in T. canis infections.

On the other hand, the effect of N. sativa extract can be partially attributed to modulation of immune response via a variety of inflammatory mediators, of which expression has been shown to be increased in response to T. canis infection.21 Many researchers have studied the effects of N. sativa or its derived thymoquinone on the immune system, and found that the extract stimulates the production of some certain cytokines, and leads to alterations in the levels of these mediators.22,23 It has been stated recently that Nigella sativa and thymoquinone have a therapeutic and protective effect by decreasing oxidative stress in streptozotocin-induced diabetic rats.3 Moreover, N. sativa has been reported to have a stimulatory effect on human lymphocytes and polymorphonuclear leukocyte phagocytic activity.24 It has also been suggested that fixed oil of N. sativa inhibits the generation of eicosanoides which play an important role in inflammation.25 We suggest that the decrease in the degree of inflammation observed in both liver and lungs of mice treated with N. sativa might be associated with this anti-eicosanoid activity.

In the present study, larval recovery levels in larval toxocariasis were higher in liver and lungs and lower in muscles and brain on day 7 after infection, which is in line with a study by Lai et al.16 This result may be attributable to proteolytic activity of enzymes produced by larvae that leads to moderate to severe histopathological changes in liver and lungs, and mild changes in the muscles and brain.

Conclusion

Our data indicate that the treatment with N. sativa at both doses or the combination dose of N. sativa and albendazole reduced the degree of inflammation and necrosis in liver, lung and brain induced by T. canis infection, lead to a reduction in the percentage of eosinophils, and markedly decreased the elevated liver enzyme levels.

Black Seed in Cuisine


Black seed is the name used for Nigella sativa, an herb in the Ranunculaceae family that produces small black seeds. Black seed is also referred to as nigella, black caraway, black cumin, and fennel flower. Recipes may use whole or crushed seeds.



United States: Black seed is an optional decorative ingredient in one version of the braided challah, a Jewish-style bread loaf served at Shabbat meals. It is commonly used in American cuisine.



Canada: Early settlers of Canada supposedly cultivated black seed extensively and used it as a substitute for nutmeg or pepper.



England: Nigella sativa was brought to England from Egypt in 1548, and its seeds were used as a seasoning.



Poland: In Polish cuisine, black seeds are primarily sprinkled on breads and rolls like poppy seeds.



India: Nigella sativa is commonly used in Indian cuisine to add flavor to curries, korma, and dahl. It is also a part of many spice mixtures, such as the Bengali five-spice, and can be found in Indian breads such as naan. It is also used in casseroles, stews and vegetable dishes.



Egypt: In ancient Egypt, Nigella sativa was sprinkled on breads and rolls. It was also mixed with other grains to make barley meal porridge, food for the afterlife.



Iran: Black seed is used to flavor vinegar, which is then utilized for pickling vegetables such as the torshi-ye leeteh and torshi-ye meeveh.



Lebanon: Black seeds are used to add flavor to cheeses, such as the Lebanese haloumi.



Turkey: A Turkish bread called corek is flavored with Nigella sativa. In most of the Middle East, the black seed is sprinkled on breads and crackers.



Ethiopia: Nigella sativa is cultivated in many provinces of Ethiopia. The seeds are used in flavoring bread and local beverages. Black seed powder is added to berbere sauce ("wot") to mask the pungency of pepper.

Black seed oil for MS

Hi all, here I would like to report my story with the BLACK SEED OIL. The oil that was capable of curing me out of Multiple Sclerosis! This fact will be followed with MRI reports that will support my point of view.

I was infected with Multiple Sclerosis that has been proven in 2001. At that time I passed through periodical intervals, lasting for 14 seconds, where I was unable to safely use any of my right side parts; Paraesthesia in my right side (Partial numbness, tingling, buzzing and vibration sensations), partial loss of sensation, pain in my right eye causing Diplopia (medical term for double vision), Shaking when performing fine movements … This act remains for three months and reached hard times passing through the stated intervals for 15 seconds every 3 minutes! And the psychiatrist told me that the MS attach was hard.

The MS attack was released after about 3 moths. But many of the symptoms remained

The previously stated intervals passing in a softer state and infecting to me in a rate of 5 to 6 times daily (taking into consideration that I’m living with no hard acts done) otherwise they’ll hit me in a larger number. And these intervals increases after 10:00 pm

Fatigue, a daily lack of energy

These symptoms remained for more than five months and the psychiatrist told me: “The MS symptoms lasting for more than 3 months after the attack are supposed to ever last; you are supposed to manage your life with these symptoms”

Actually I tried to take several types of vitamins to enable me continue my study at university with no use! Actually I was so depressed; I want to continue my life and take my BA in Architectural Engineering but I can’t! I have to submit my late projects that were affected by the period I was passing the MS attack and I have to continue studying

After that I was attending the pharmacy exhibition in our university, and what had caught my attention a project done by a group of First Year Pharmacy about the BLACK SEED OIL

Several facts about the BLACK SEED OIL stated in the project stucked in my mind

The increase in the immunity by 77% for the users of the BLACK SEED OIL. Although I knew that the MS is SUPPOSED to be auto immune, BUT I believed that the BLACK SEED OIL will fix that trouble in the immunity system

The proven ability of the BLACK SEED OIL in providing the body with power. The power that I had lost out of MS and I was in a big need for it

I start taking daily, and orally a table spoon of the black seed oil with a cup of orange juice after it directly; due to the fact that the taste of the BLACK SEED OIL is somehow hard

And here was the shock! After taking this recipe for three days, I was capable of studying up to 2pm! And after 15days I got rid of all the MS symptoms that were not solved by any medication

This sudden change encourages me to search the net about the BLACK SEED OIL where I found fantastic results. You can do that. The most important fact I found is the one stating that the BLACK SEED OIL encourages the body for the production of Interferon.

I remained in taking a tablespoon of the BLACK SEED OIL daily for around 1.5 years although I retained my power after 15 days. But as you can say, the fantastic results I found encourage me on remaining in taking the BLACK SEED OIL as a kind of protection

In the following lines, I will quote words of my MRI reports examinations;

- FIRST MRI EXAMINATION (17-03-2003): There are multiple rounded and oval-shaped lesions ranging from 2mm to 10mm in size involving the corpus callosum. This MRI examination in addition to other examination (mainly the liquid of the stamp) proves my infection with Multiple Sclerosis.





- SECOND MRI EXAMINATION. This examination is done after the first examination in one year. Actually it had proven that the lesions in brain have increased! It was a sudden to me because I was expecting that the BLACK SEED OIL will stop the growth of lesions. But I remained on taking the BLACK SEED OIL; at least it was giving me power.





- THIRD MRI EXAMINATION (12-10-2005): and here was the nice surprise

The ventricular system is not enlarged.

No effect is seen.

No abnormal nodular enhancing lesion is seen.

The cranio cervical junction and the CP are preserved.

The examination proves that the lesions are not increased in size after 2.5 years. More than that, the psychiatrist told me that this examination, compared to the previous one, proves that the lesions in brain are decreased in size. And that is impossible to happen. He told me: “This examination leads me to be suspect with everything I have studied in my life”

At that time, I’ve concluded an important fact; in the second MRI examination that proves the increase of lesions, I have done it after taking the BLACK SEED OIL in 4 months. But there were 8 months in which the multiple sclerosis was acting actively. And case I haven’t taken the BLACK SEED OIL, sure I could examined another MS attack.

- FOURTH MRI EXAMINATION (13-07-2010): After seven years, and upon the request of several members that asked me to do a new MRI examination just as a kind of test, I’ve done an MRI examination and here are some of its states facts;

Normal size and shape of the ventricular system.

No shift of midline structures.

No evidence of intra cerebral bleeding or extra axial collections.

Normal height, signal characteristics and enhancement of pituitary gand. Central pituitary stalk.

No sellar or parasellar masses.

The psychiatrist told me: “The MS in your case is completely stopped. Even in my bed examination to you, nothing appeared.

From this story, I’d like to state several points;

I have been living till now, with the proof of the MRI examination for 2.5+5=7.5 years with no MS attack.

The second MRI examination proves that the MS was active and the BLACK SEED OIL has stopped its growth.

In all these years, I haven’t taken any of the proposed medications of the interferon injections.

The BLACK SEED OIL has no side effects. You can check that by a simple yahoo / Google search. Case it hadn’t benefit you (I don’t feel that!) it will not cause you any harm
